Uncertain significance — the classification assigned by Women's Health and Genetics/Laboratory Corporation of America, LabCorp to NM_001009944.3(PKD1):c.9202-3C>G, citing LabCorp Variant Classification Summary - May 2015: Variant summary: PKD1 c.9202-3C>G alters a non-conserved nucleotide located close to a canonical splice site and therefore could affect mRNA splicing, leading to a significantly altered protein sequence. Several computational tools predict a significant impact on normal splicing: Three predict the variant abolishes a canonical 3' acceptor site, two predict the variant weakens a canonical 3' acceptor site, and four predict the variant creates a cryptic 3' acceptor site. However, these predictions have yet to be confirmed by functional studies. The frequency data for this variant in gnomAD is considered unreliable, as metrics indicate poor data quality at this position. c.9202-3C>G has been observed in at least one individual affected with polycystic kidney disease (example: Simms_2015). This report does not provide unequivocal conclusions about association of the variant with PKD1-related conditions. To our knowledge, no experimental evidence demonstrating an impact on protein function has been reported.
