This variant lies in the TP53 gene (transcript NM_000546.6) at coding-DNA position 884, where C is replaced by A; at the protein level this means replaces proline at residue 295 with histidine — a missense variant. Submitter rationale: Variant summary: TP53 c.884C>A (p.Pro295His) results in a non-conservative amino acid change in the encoded protein sequence. Three of five in-silico tools predict a benign effect of the variant on protein function. The variant was absent in 251492 control chromosomes (gnomAD). The available data on variant occurrences in the general population are insufficient to allow any conclusion about variant significance. To our knowledge, no occurrence of c.884C>A in individuals affected with Li-Fraumeni Syndrome has been reported. The IARC TP53 database reports this variant to be non-functional based on transcriptional activity in yeast (Kato_2003). However, a loss-of-function saturation mutagenesis screen indicated that this missense does not substantially affect TP53 function (Giacomelli_2018). Therefore, these reports do not allow convincing conclusions about the variant effect. The following publications have been ascertained in the context of this evaluation (PMID: 12826609, 30224644).
