NM_000104.4(CYP1B1):c.1195A>G (p.Ile399Val) was classified as Uncertain significance by Women's Health and Genetics/Laboratory Corporation of America, LabCorp, citing LabCorp Variant Classification Summary - May 2015: Variant summary: CYP1B1 c.1195A>G (p.Ile399Val) results in a conservative amino acid change in the encoded protein sequence. Three of five in-silico tools predict a damaging effect of the variant on protein function. The variant allele was found at a frequency of 1.2e-05 in 251118 control chromosomes. This variant was also observed in the homozygous state in 1 control individual (example, Chitsazian_2007). The available data on variant occurrences in the general population are insufficient to allow any conclusion about variant significance. To our knowledge, no occurrence of c.1195A>G in individuals affected with Primary Congenital Glaucoma and no experimental evidence demonstrating its impact on protein function have been reported.
