Submitter rationale: Variant summary: ATP7B c.2954G>A (p.Cys985Tyr) results in a non-conservative amino acid change in the encoded protein sequence. Five of five in-silico tools predict a damaging effect of the variant on protein function. The variant was absent in 249270 control chromosomes. c.2954G>A has been reported in the literature in individuals affected with Wilson Disease in either the compound heterozygous state or heterozygous without a second allele specified (Wiernicka_2013, Haas_1999, Kluska_2019, Pham_2017). These data indicate that the variant may be associated with disease. To our knowledge, no experimental evidence demonstrating an impact on protein function has been reported.
