Uncertain significance — the classification assigned by Women's Health and Genetics/Laboratory Corporation of America, LabCorp to NM_022081.6(HPS4):c.1856C>T (p.Pro619Leu), citing LabCorp Variant Classification Summary - May 2015: Variant summary: HPS4 c.1856C>T (p.Pro619Leu) results in a non-conservative amino acid change located in the CCZ1/INTU/HPS4, third Longin domain (IPR043989) of the encoded protein sequence. Four of five in-silico tools predict a damaging effect of the variant on protein function. The variant allele was found at a frequency of 2e-05 in 249614 control chromosomes. c.1856C>T has been reported in the literature in individuals affected with Albinism (Lasseaux_2018). These data indicate that the variant may be associated with disease. To our knowledge, no experimental evidence demonstrating an impact on protein function has been reported.
