Pathogenic for Joubert syndrome and related disorders — the classification assigned by Women's Health and Genetics/Laboratory Corporation of America, LabCorp to NM_153704.6(TMEM67):c.274G>A (p.Gly92Arg), citing LabCorp Variant Classification Summary - May 2015: Variant summary: TMEM67 c.274G>A (p.Gly92Arg) results in a non-conservative amino acid change in the encoded protein sequence. Four of five in-silico tools predict a damaging effect of the variant on protein function. The variant allele was found at a frequency of 1.6e-05 in 246784 control chromosomes. c.274G>A has been reported in the literature in the homozygous and compound heterozygous states multiple individuals affected with Joubert Syndrome And Related Disorders (e.g. Szymanska_20012, Park_2016, Konig_2022). These data indicate that the variant is very likely to be associated with disease. To our knowledge, no experimental evidence demonstrating an impact on protein function has been reported.
