NM_004608.4(TBX6):c.990del (p.Glu332fs) was classified as Pathogenic for Spondylocostal dysostosis 5 by Women's Health and Genetics/Laboratory Corporation of America, LabCorp, citing LabCorp Variant Classification Summary - May 2015: Variant summary: TBX6 c.990delC (p.Glu332LysfsX166) causes a frameshift which results in an extension of the protein. Two downtream protein extension variants ending at the same termination codon have been reported as associated with disease (c.1061delC p.Ala354GlyfsX144 and c.994delG p.Glu332LysfsX166, both of which was reported in trans with a hypomorphic haplotype T-C-A, PMID 36161696, 35726512).The current variant was absent in 222692 control chromosomes. To our knowledge, no occurrence of c.990delC in individuals affected with Spondylocostal Dysostosis 5 and no experimental evidence demonstrating its impact on protein function have been reported. No submitters have cited clinical-significance assessments for this variant to ClinVar. Based on the evidence outlined above, the variant was classified as pathogenic.
