Uncertain significance — the classification assigned by Women's Health and Genetics/Laboratory Corporation of America, LabCorp to NM_000101.4(CYBA):c.371C>G (p.Ala124Gly), citing LabCorp Variant Classification Summary - May 2015: Variant summary: CYBA c.371C>G (p.Ala124Gly) results in a non-conservative amino acid change in the encoded protein sequence. Five of five in-silico tools predict a damaging effect of the variant on protein function. Consensus agreement among computation tools predict no significant impact on normal splicing. However, these predictions have yet to be confirmed by functional studies. The variant was absent in 129404 control chromosomes (gnomAD). The available data on variant occurrences in the general population are insufficient to allow any conclusion about variant significance. To our knowledge, no occurrence of c.371C>G in individuals affected with Chronic Granulomatous Disease and no experimental evidence demonstrating its impact on protein function have been reported. No submitters have cited clinical-significance assessments for this variant to ClinVar. However, another variant at the same codon, p.Ala124Val, has been classified as pathogenic/likely pathogenic by our lab and other ClinVar submitters (Variation ID: 68212). Based on the evidence outlined above, the variant was classified as uncertain significance.
