Pathogenic for Marfan Syndrome/Loeys-Dietz Syndrome/Familial Thoracic Aortic Aneurysms and Dissections — the classification assigned by Women's Health and Genetics/Laboratory Corporation of America, LabCorp to NM_000138.5(FBN1):c.4755del (p.Glu1584_Tyr1585insTer), citing LabCorp Variant Classification Summary - May 2015: Variant summary: FBN1 c.4755delC (p.Tyr1585X) results in a premature termination codon, predicted to cause a truncation of the encoded protein or absence of the protein due to nonsense mediated decay, which are commonly known mechanisms for disease. The variant was absent in 250650 control chromosomes (gnomAD). c.4755delC has been reported in the literature in at least an individual affected with clinical features of Marfan Syndrome (example: Baudhuin_2015). To our knowledge, no experimental evidence demonstrating an impact on protein function has been reported. The following publication has been ascertained in the context of this evaluation (PMID: 25101912). No submitters have cited clinical-significance assessments for this variant to ClinVar. Based on the evidence outlined above, the variant was classified as pathogenic.
