This variant lies in the TTN gene (transcript NM_001267550.2) at 2263 bases into the intron immediately after coding-DNA position 11311, duplicating one base. Submitter rationale: This variant is present in the healthy population databases with low frequency (gnomAD v4.1.0). This variant induces a frameshift in Novex-3 transcript with protein truncation. However, the variant is not present in the major cardiac transcripts (N2BA, N2B). Novex-3 transcript might be relevant for fetal cardiomyocytes division and its interaction with lamin A/B during the same developmental period. In adult life, the proportion of Novex-3 transcripts is estimated <10% of the total number of cardiac TTN transcripts. Therefore, the variant was classified as uncertain, according to ACMG 2015 and ClinGen criteria.
