Submitter rationale: Variant summary: SPG7 c.2081G>A (p.Gly694Asp) results in a non-conservative amino acid change located in the Peptidase M41 domain (IPR000642) of the encoded protein sequence. Four of five in-silico tools predict a damaging effect of the variant on protein function. The frequency data for this variant in gnomAD is considered unreliable, as metrics indicate poor data quality at this position. c.2081G>A has been reported in the literature in at least one compound heterozygous individual affected with Hereditary Spastic Paraplegia 7 (Roxburgh_2013). These report(s) do not provide unequivocal conclusions about association of the variant with Hereditary Spastic Paraplegia 7. To our knowledge, no experimental evidence demonstrating an impact on protein function has been reported.
