Uncertain significance — the classification assigned by Women's Health and Genetics/Laboratory Corporation of America, LabCorp to NM_000132.4(F8):c.5924T>C (p.Ile1975Thr), citing LabCorp Variant Classification Summary - May 2015. This variant lies in the F8 gene (transcript NM_000132.4) at coding-DNA position 5924, where T is replaced by C; at the protein level this means replaces isoleucine at residue 1975 with threonine — a missense variant. Submitter rationale: Variant summary: F8 c.5924T>C (p.Ile1975Thr) results in a non-conservative amino acid change located in the Multicopper oxidase, C-terminal domain (IPR011706) of the encoded protein sequence. Five of five in-silico tools predict a damaging effect of the variant on protein function. The variant was absent in 183283 control chromosomes (gnomAD). c.5924T>C has been reported in the literature in individuals affected with Factor VIII Deficiency (Hemophilia A; Cutler_2002, Green_2008, Johnsen_2017, EAHAD F8 Variant Database). These data indicate that the variant may be associated with disease. To our knowledge, no experimental evidence demonstrating an impact on protein function has been reported. The following publications have been ascertained in the context of this evaluation (PMID: 11857744, 18691168, 29296726). No submitters have cited clinical-significance assessments for this variant to ClinVar. Based on the evidence outlined above, the variant was classified as VUS-possibly pathogenic.
