A presumed nomenclature of c.(53+1_54-1)_(*1977_?)dup has been designated for the purposes of this classification. The exact breakpoint at the 3' end of this variant is unknown, therefore this duplication may extend downstream of the annotated region of the gene. As it duplicates the termination codon, its effect on the encoded protein is unknown. A large duplication which encompasses exons 2-5 in the ZC4H2 gene (Size: ~61 kbp) was found at a frequency of 6.3e-05 in 15814 control chromosomes in the gnomAD database (Structural Variants v2.1 dataset). The available data on variant occurrences in the general population are insufficient to allow any conclusion about variant significance. To our knowledge, no occurrence of c.(53+1_54-1)_(*1977_?)dup in individuals affected with Wieacker-Wolff Syndrome and no experimental evidence demonstrating its impact on protein function have been reported.
