NM_003172.4(SURF1):c.23A>T (p.Gln8Leu) was classified as Uncertain significance by Women's Health and Genetics/Laboratory Corporation of America, LabCorp, citing LabCorp Variant Classification Summary - May 2015: Variant summary: SURF1 c.23A>T (p.Gln8Leu) results in a non-conservative amino acid change in the encoded protein sequence. Four of five in-silico tools predict a benign effect of the variant on protein function. The variant was absent in 56782 control chromosomes. The available data on variant occurrences in the general population are insufficient to allow any conclusion about variant significance. c.23A>T has been reported in the literature without a clear genotype/zygosity specification in at-least one individual affected with Leigh Syndrome (example, Diogo_2009 cited in Li_2018). These report(s) do not provide unequivocal conclusions about association of the variant with Leigh Syndrome. To our knowledge, no experimental evidence demonstrating an impact on protein function has been reported. The following publications have been ascertained in the context of this evaluation (PMID: 19380071, 29933018). No submitters have cited clinical-significance assessments for this variant to ClinVar. Based on the evidence outlined above, the variant was classified as uncertain significance.
