Uncertain significance — the classification assigned by Women's Health and Genetics/Laboratory Corporation of America, LabCorp to NM_005055.5(RAPSN):c.1172T>C (p.Leu391Pro), citing LabCorp Variant Classification Summary - May 2015: Variant summary: RAPSN c.1172T>C (p.Leu391Pro) results in a non-conservative amino acid change located in the Zinc finger, RING-type domain (IPR001841) of the encoded protein sequence. Five of five in-silico tools predict a damaging effect of the variant on protein function. The variant was absent in 155740 control chromosomes. c.1172T>C has been reported in the literature in compound heterozygous individuals affected with RAPSN-Related Disorders (Petrovski_2019). These data indicate that the variant may be associated with disease. To our knowledge, no experimental evidence demonstrating an impact on protein function has been reported.
