Uncertain significance — the classification assigned by Women's Health and Genetics/Laboratory Corporation of America, LabCorp to NM_004937.3(CTNS):c.615C>A (p.Asp205Glu), citing LabCorp Variant Classification Summary - May 2015: Variant summary: CTNS c.615C>A (p.Asp205Glu) results in a conservative amino acid change in the encoded protein sequence. Four of five in-silico tools predict a damaging effect of the variant on protein function. The variant was absent in 251346 control chromosomes. The available data on variant occurrences in the general population are insufficient to allow any conclusion about variant significance. To our knowledge, no occurrence of c.615C>A in individuals affected with Cystinosis and no experimental evidence demonstrating its impact on protein function have been reported. A different variant affecting the same codon has been classified as pathogenic by our lab (c.613G>A, Asp205Asn), supporting the critical relevance of codon 205 to CTNS protein function, however additional evidence is needed for this variant to allow conclusions about variant significance. No submitters have cited clinical-significance assessments for this variant to ClinVar. Based on the evidence outlined above, the variant was classified as uncertain significance.
