This variant lies in the WDR35 gene (transcript NM_020779.4) at coding-DNA position 1871, where T is replaced by G; at the protein level this means replaces isoleucine at residue 624 with serine — a missense variant. Submitter rationale: The I635S variant in the WDR35 gene gene has not been reported previously as a pathogenic variant, nor as a benign variant, to our knowledge. The I635S variant is observed in 8/9692 (0.08%) alleles from individuals of South Asian background, in large population cohorts (Lek et al., 2016; 1000 Genomes Consortium et al., 2015; Exome Variant Server). The I635S variant is a non-conservative amino acid substitution, which is likely to impact secondary protein structure as these residues differ in polarity, charge, size and/or other properties. This substitution occurs at a position that is conserved across species. In silico analysis predicts this variant is probably damaging to the protein structure/function. We interpret I635S as a variant of uncertain significance.
