Uncertain significance for Inborn genetic diseases — the classification assigned by Ambry Genetics to NM_001184.4(ATR):c.22C>G (p.Leu8Val), citing Ambry Variant Classification Scheme 2023: The p.L8V variant (also known as c.22C>G), located in coding exon 1 of the ATR gene, results from a C to G substitution at nucleotide position 22. The leucine at codon 8 is replaced by valine, an amino acid with highly similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
