Uncertain significance for Familial thoracic aortic aneurysm and aortic dissection — the classification assigned by Ambry Genetics to NM_000393.5(COL5A2):c.3341G>A (p.Arg1114Gln), citing Ambry Variant Classification Scheme 2023: The p.R1114Q variant (also known as c.3341G>A), located in coding exon 47 of the COL5A2 gene, results from a G to A substitution at nucleotide position 3341. The arginine at codon 1114 is replaced by glutamine, an amino acid with highly similar properties. This amino acid position is not well conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
