Uncertain significance for Familial thoracic aortic aneurysm and aortic dissection — the classification assigned by Ambry Genetics to NM_000090.4(COL3A1):c.3998A>G (p.Asp1333Gly), citing Ambry Variant Classification Scheme 2023: The p.D1333G variant (also known as c.3998A>G), located in coding exon 49 of the COL3A1 gene, results from an A to G substitution at nucleotide position 3998. The aspartic acid at codon 1333 is replaced by glycine, an amino acid with similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
