Submitter rationale: The p.V892F variant (also known as c.2674G>T), located in coding exon 23 of the TSC2 gene, results from a G to T substitution at nucleotide position 2674. The valine at codon 892 is replaced by phenylalanine, an amino acid with highly similar properties. This alteration was identified in 1 of 374 patients with clinically suspected TSC undergoing genetic testing within the TSC1 and TSC2 genes (Meng Y et al. J Hum Genet, 2021 Mar;66:227-236). This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be deleterious by in silico analysis.
