This variant lies in the TSC2 gene (transcript NM_000548.5) at the canonical splice acceptor site of the intron immediately before coding-DNA position 3285, where G is replaced by C; at the protein level this means a change at this position may disrupt normal splicing. Submitter rationale: The c.3285-1G>C intronic variant results from a G to C substitution one nucleotide upstream from coding exon 28 of the TSC2 gene. Alterations that disrupt the canonical splice site are expected to result in aberrant splicing. This nucleotide position is highly conserved in available vertebrate species. In silico splice site analysis predicts that this alteration will weaken the native splice acceptor site and may result in the creation or strengthening of a novel splice acceptor site. One possible resulting transcript is predicted to be in-frame and is not expected to trigger nonsense-mediated mRNA decay. The exact functional effect of the altered amino acid sequence is unknown.
