NM_000368.5(TSC1):c.1016C>G (p.Thr339Ser) was classified as Uncertain significance for Hereditary cancer-predisposing syndrome by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.T339S variant (also known as c.1016C>G), located in coding exon 8 of the TSC1 gene, results from a C to G substitution at nucleotide position 1016. The threonine at codon 339 is replaced by serine, an amino acid with similar properties. This amino acid position is conserved. In addition, the in silico prediction for this alteration is inconclusive. Based on the available evidence, the clinical significance of this variant remains unclear.
