NM_017636.4(TRPM4):c.3637A>G (p.Lys1213Glu) was classified as Uncertain significance for Cardiovascular phenotype by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.K1213E variant (also known as c.3637A>G), located in coding exon 24 of the TRPM4 gene, results from an A to G substitution at nucleotide position 3637. The lysine at codon 1213 is replaced by glutamic acid, an amino acid with similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
