NM_017636.4(TRPM4):c.2794TTCTTCCTC[3] (p.Leu937_Gly938insPhePheLeu) was classified as Uncertain significance for Cardiovascular phenotype by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The c.2803_2811dupTTCTTCCTC variant (also known as p.F935_L937dup), located in coding exon 19 of the TRPM4 gene, results from an in-frame duplication of TTCTTCCTC at nucleotide positions 2803 to 2811. This results in the duplication of 3 extra residues (FFL) between codons 935 and 937. This amino acid region ranges from highly conserved to well conserved in available vertebrate species. In addition, this alteration is predicted to be deleterious by in silico analysis (Choi Y et al. PLoS ONE. 2012; 7(10):e46688). Based on the available evidence, the clinical significance of this variant remains unclear.
