Uncertain significance — the classification assigned by Ambry Genetics to NM_018975.4(TERF2IP):c.634A>C (p.Lys212Gln), citing Ambry Variant Classification Scheme 2023: The p.K212Q variant (also known as c.634A>C), located in coding exon 1 of the TERF2IP gene, results from an A to C substitution at nucleotide position 634. The lysine at codon 212 is replaced by glutamine, an amino acid with similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
