Uncertain significance for Cardiovascular phenotype — the classification assigned by Ambry Genetics to NM_181486.4(TBX5):c.991T>G (p.Cys331Gly), citing Ambry Variant Classification Scheme 2023: The p.C331G variant (also known as c.991T>G), located in coding exon 8 of the TBX5 gene, results from a T to G substitution at nucleotide position 991. The cysteine at codon 331 is replaced by glycine, an amino acid with highly dissimilar properties. This amino acid position is conserved. In addition, this alteration is predicted to be deleterious by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
