Pathogenic for Cardiovascular phenotype — the classification assigned by Ambry Genetics to NM_000335.5(SCN5A):c.2595C>G (p.Tyr865Ter), citing Ambry Variant Classification Scheme 2023: The p.Y865* pathogenic mutation (also known as c.2595C>G), located in coding exon 15 of the SCN5A gene, results from a C to G substitution at nucleotide position 2595. This changes the amino acid from a tyrosine to a stop codon within coding exon 15. This variant is considered to be rare based on population cohorts in the Genome Aggregation Database (gnomAD). This alteration is expected to result in loss of function by premature protein truncation or nonsense-mediated mRNA decay. As such, this alteration is interpreted as a disease-causing mutation.
