Uncertain significance — the classification assigned by Ambry Genetics to NM_002907.4(RECQL):c.304G>T (p.Val102Leu), citing Ambry Variant Classification Scheme 2023: The p.V102L variant (also known as c.304G>T), located in coding exon 3 of the RECQL gene, results from a G to T substitution at nucleotide position 304. The valine at codon 102 is replaced by leucine, an amino acid with highly similar properties. This amino acid position is not well conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. The evidence for this gene-disease relationship is limited; therefore, the clinical significance of this alteration is unclear.
