This variant lies in the PTEN gene (transcript NM_000314.8) at coding-DNA position 618, where C is replaced by G; at the protein level this means replaces phenylalanine at residue 206 with leucine — a missense variant. Submitter rationale: The p.F206L variant (also known as c.618C>G), located in coding exon 6 of the PTEN gene, results from a C to G substitution at nucleotide position 618. The phenylalanine at codon 206 is replaced by leucine, an amino acid with highly similar properties. This variant demonstrated possibly low intracellular protein abundance in a massively parallel functional assay (Matreyek KA et al. Nat Genet, 2018 Jun;50:874-882).
