Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_002691.4(POLD1):c.399G>T (p.Glu133Asp), citing Ambry Variant Classification Scheme 2023: The p.E133D variant (also known as c.399G>T), located in coding exon 3 of the POLD1 gene, results from a G to T substitution at nucleotide position 399. The glutamic acid at codon 133 is replaced by aspartic acid, an amino acid with highly similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
