Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_000535.7(PMS2):c.2028G>A (p.Met676Ile), citing Ambry Variant Classification Scheme 2023: The p.M676I variant (also known as c.2028G>A), located in coding exon 12 of the PMS2 gene, results from a G to A substitution at nucleotide position 2028. The methionine at codon 676 is replaced by isoleucine, an amino acid with highly similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be deleterious by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
