The variant was absent in 250954 control chromosomes (gnomAD). c.247A>G has been reported in the literature in individuals affected with Autoimmune Polyglandular Syndrome Type 1 (e.g. Nagamine_1997, Perheentupa_2006, Meisel_2021). These data indicate that the variant may be associated with disease. At least one publication reported experimental evidence evaluating an impact on protein function, and demonstrated subcellular mislocalization pattern, with decreased homomultimerization and transactivation capacity (Halonen_2004).
