Uncertain significance — the classification assigned by Ambry Genetics to NM_001166108.2(PALLD):c.734A>G (p.Asp245Gly), citing Ambry Variant Classification Scheme 2023: The p.D245G variant (also known as c.734A>G), located in coding exon 1 of the PALLD gene, results from an A to G substitution at nucleotide position 734. The aspartic acid at codon 245 is replaced by glycine, an amino acid with similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
