Uncertain significance — the classification assigned by Ambry Genetics to NM_001386125.1(OBSCN):c.18871T>C (p.Tyr6291His), citing Ambry Variant Classification Scheme 2023: The p.Y5334H variant (also known as c.16000T>C), located in coding exon 59 of the OBSCN gene, results from a T to C substitution at nucleotide position 16000. The tyrosine at codon 5334 is replaced by histidine, an amino acid with similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Based on the available evidence, the clinical significance of this alteration remains unclear.
