Uncertain significance — the classification assigned by Ambry Genetics to NM_002519.3(NPAT):c.3275C>G (p.Ala1092Gly), citing Ambry Variant Classification Scheme 2023: The p.A1092G variant (also known as c.3275C>G), located in coding exon 17 of the NPAT gene, results from a C to G substitution at nucleotide position 3275. The alanine at codon 1092 is replaced by glycine, an amino acid with similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
