Uncertain significance for Familial thoracic aortic aneurysm and aortic dissection — the classification assigned by Ambry Genetics to NM_017617.5(NOTCH1):c.5764A>G (p.Asn1922Asp), citing Ambry Variant Classification Scheme 2023: The p.N1922D variant (also known as c.5764A>G), located in coding exon 31 of the NOTCH1 gene, results from an A to G substitution at nucleotide position 5764. The asparagine at codon 1922 is replaced by aspartic acid, an amino acid with highly similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Based on the available evidence, the clinical significance of this alteration remains unclear.
