NM_003491.4(NAA10):c.154A>G (p.Ile52Val) was classified as Uncertain significance for Inborn genetic diseases by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The c.154A>G (p.I52V) alteration is located in exon 3 (coding exon 3) of the NAA10 gene. This alteration results from a A to G substitution at nucleotide position 154, causing the isoleucine (I) at amino acid position 52 to be replaced by a valine (V). This variant was not reported in population-based cohorts in the Genome Aggregation Database (gnomAD). This amino acid position is highly conserved in available vertebrate species. This missense alteration is located in a region that has a low rate of benign missense variation (Lek, 2016; Firth, 2009). This alteration is predicted to be tolerated by in silico analysis; however, in silico splice site analysis predicts that this alteration may weaken the native splice donor site. Based on insufficient or conflicting evidence, the clinical significance of this alteration remains unclear.
