Uncertain significance for Familial thoracic aortic aneurysm and aortic dissection — the classification assigned by Ambry Genetics to NM_053025.4(MYLK):c.5707G>C (p.Glu1903Gln), citing Ambry Variant Classification Scheme 2023: The p.E1903Q variant (also known as c.5707G>C), located in coding exon 31 of the MYLK gene, results from a G to C substitution at nucleotide position 5707. The glutamic acid at codon 1903 is replaced by glutamine, an amino acid with highly similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
