NM_053025.4(MYLK):c.3099G>C (p.Glu1033Asp) was classified as Uncertain significance for Familial thoracic aortic aneurysm and aortic dissection by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.E1033D variant (also known as c.3099G>C), located in coding exon 15 of the MYLK gene, results from a G to C substitution at nucleotide position 3099. The glutamic acid at codon 1033 is replaced by aspartic acid, an amino acid with highly similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
