Uncertain significance for Inborn genetic diseases — the classification assigned by Ambry Genetics to NM_015057.5(MYCBP2):c.9193C>T (p.His3065Tyr), citing Ambry Variant Classification Scheme 2023: The c.9193C>T (p.H3065Y) alteration is located in coding exon 56 of the MYCBP2 gene. This alteration results from a C to T substitution at nucleotide position 9193, causing the histidine (H) at amino acid position 3065 to be replaced by a tyrosine (Y). This variant was not reported in population-based cohorts in the Genome Aggregation Database (gnomAD). This amino acid position is highly conserved in available vertebrate species. The in silico prediction for this alteration is inconclusive. Based on insufficient or conflicting evidence, the clinical significance of this alteration remains unclear.
