Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_000179.3(MSH6):c.3386G>C (p.Cys1129Ser), citing Ambry Variant Classification Scheme 2023: The p.C1129S variant (also known as c.3386G>C), located in coding exon 5 of the MSH6 gene, results from a G to C substitution at nucleotide position 3386. The cysteine at codon 1129 is replaced by serine, an amino acid with dissimilar properties. This amino acid position is conserved. In addition, this alteration is predicted to be deleterious by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
