Uncertain significance — the classification assigned by Ambry Genetics to NM_002432.3(MNDA):c.398C>T (p.Ala133Val), citing Ambry Variant Classification Scheme 2023: The p.A133V variant (also known as c.398C>T), located in coding exon 2 of the MNDA gene, results from a C to T substitution at nucleotide position 398. The alanine at codon 133 is replaced by valine, an amino acid with similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
