Uncertain significance for Inborn genetic diseases — the classification assigned by Ambry Genetics to NM_020774.4(MIB1):c.252G>A (p.Met84Ile), citing Ambry Variant Classification Scheme 2023: The p.M84I variant (also known as c.252G>A), located in coding exon 2 of the MIB1 gene, results from a G to A substitution at nucleotide position 252. The methionine at codon 84 is replaced by isoleucine, an amino acid with highly similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be deleterious by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
