Several computational tools predict a significant impact on normal splicing: One predict the variant no significant impact on splicing. Two predict the variant weakens a 5' donor site. One predict the variant abolishes a 5' splicing donor site. However, these predictions have yet to be confirmed by functional studies. The variant allele was found at a frequency of 8.4e-05 in 251410 control chromosomes. This frequency is not significantly higher than estimated for a pathogenic variant in RYR1 causing Congenital multicore myopathy with external ophthalmoplegia (8.4e-05 vs 0.0011), allowing no conclusion about variant significance. To our knowledge, no occurrence of c.3555C>T in individuals affected with Congenital multicore myopathy with external ophthalmoplegia and no experimental evidence demonstrating its impact on protein function have been reported.
