NM_052947.4(ALPK2):c.1805G>A (p.Cys602Tyr) was classified as Uncertain significance by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.C602Y variant (also known as c.1805G>A), located in coding exon 3 of the ALPK2 gene, results from a G to A substitution at nucleotide position 1805. The cysteine at codon 602 is replaced by tyrosine, an amino acid with highly dissimilar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Based on the available evidence, the clinical significance of this alteration remains unclear.
