Uncertain significance — the classification assigned by Ambry Genetics to NM_052947.4(ALPK2):c.6066C>A (p.Asn2022Lys), citing Ambry Variant Classification Scheme 2023: The p.N2022K variant (also known as c.6066C>A), located in coding exon 10 of the ALPK2 gene, results from a C to A substitution at nucleotide position 6066. The asparagine at codon 2022 is replaced by lysine, an amino acid with similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Based on the available evidence, the clinical significance of this alteration remains unclear.
