Uncertain significance — the classification assigned by Ambry Genetics to NM_001365951.3(KIF1B):c.3149C>G (p.Ala1050Gly), citing Ambry Variant Classification Scheme 2023: The p.A1004G variant (also known as c.3011C>G), located in coding exon 27 of the KIF1B gene, results from a C to G substitution at nucleotide position 3011. The alanine at codon 1004 is replaced by glycine, an amino acid with similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
