Uncertain significance — the classification assigned by Ambry Genetics to NM_001365951.3(KIF1B):c.2870G>C (p.Ser957Thr), citing Ambry Variant Classification Scheme 2023: The p.S911T variant (also known as c.2732G>C), located in coding exon 24 of the KIF1B gene, results from a G to C substitution at nucleotide position 2732. The serine at codon 911 is replaced by threonine, an amino acid with similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
