Uncertain significance — the classification assigned by Ambry Genetics to NM_001365951.3(KIF1B):c.342C>A (p.Ser114Arg), citing Ambry Variant Classification Scheme 2023: The p.S114R variant (also known as c.342C>A), located in coding exon 3 of the KIF1B gene, results from a C to A substitution at nucleotide position 342. The serine at codon 114 is replaced by arginine, an amino acid with dissimilar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
